SCF对高糖环境下豚鼠膀胱Cajal样间质细胞超微结构的影响

摘要 目的探讨干细胞因子(SCF)对高糖环境下豚鼠膀胱Cajal样间质细胞(ICCs-like)超微结构的影响。方法胶原酶消化法原代分离培养豚鼠膀胱ICCs样细胞。实验分组:正常对照组(葡萄糖浓度为5 mmol/L)、高糖组(葡萄糖浓度为15 mmol/L),培养基培养72 h后高糖组分为高糖对照组、SCF 50 ng/ml组、SCF 100 ng/ml组,继续培养24 h及72 h。透射电镜观察各组不同时间段ICCs样细胞超微结构。结果随着培养时间延长,正常对照组ICCs样细胞内超微结构无明显变化;高糖对照组ICCs样细胞内细胞器明显减少,线粒体肿胀、大小不等、空泡样变甚至溶解;内质网扩张,胞质广泛溶解,胞周突起变短乃至消失;而SCF组ICCs样细胞超微结构得到逐步改善,细胞器增多、形态趋于正常对照组,胞周突起增多。结论外源性SCF干预后对高糖环境造成的ICCs样细胞超微结构破坏有一定的修复或逆转作用。 Purpose To investigate the effects of stem cell factor(SCF)on the ultrastructure of interstitial cells of Cajal-like cells(ICCs-like) in the bladder of guinea pig in the high glucose environment.Methods The ICCs-like cells was isolated from the bladder of guinea pig by collagenase digestion,and divided into normal control group(glucose concentration 5 mmol/L)and high glucose group(glucose concentration 15 mmol/L).After 72 h culture,the high glucose group was divided into high glucose control group,SCF 50 ng/ml group and SCF 100 ng/ml group,continued to cultivate for 24 h and 72 h.The ultrastructural changes of ICCs-like cells in groups and different time were observed by transmission electron microscope.Results With the increase of culture time,ICCs-like cells in normal control group,no significant changes were observed in the ultrastructure.Compared to the normal control group,the major change of ICCs-like cells in high glucose control group showed as follows:the organelles were decreased;mitochondrion was swelling,inequality in size,vacuoles and even dissolving,endoplasmic reticulum was extended,cytoplasm was dissolving,vacuoles were formed,the scabrosity became shorter or disappeared.SCF groups gradually tended to the normal control group.Conclusion Exogenous SCF may improve the ultrastructural damage of ICCs-like cells in the high glucose environment.
