Expression and Significance of PDGF-D and Its Receptor in Gastric Carcinoma Tissues

摘要 目的探讨血小板衍生生长因子-D(PDGF-D)及其受体PDGFRβ mRNA在胃癌组织中的表达及其与临床病理特征的关系。方法用RT-PCR方法检测58例胃癌及相对应的癌旁标本中PDGF-D及PDGFRβ mRNA的表达,分析其与临床病理特征的关系。结果 PDGF-D及PDGFRβ在胃癌组织中的阳性表达率(98.28%,72.41%)均高于癌旁组织(87.93%,53.45%),P均<0.05。PDGFRβ与PDGF-D的表达呈正相关(r=0.40,P<0.05)。PDGF-D mRNA的表达水平与胃癌的淋巴结及远处转移和复发有关(P<0.05),与其他临床病理特征无关(P>0.05)。结论 PDGF-D在胃癌的发生、发展和转移、复发中可能起着重要的作用,可作为预测胃癌转移和复发的有意义指标。 Objective To investigate the correlation between mRNA expression of PDGF-D and PDGFRβ and clinicopathological features in gastric cancer.Methods The reverse transcription-polymerase chain reaction(RT-PCR)technique was used to examine mRNA expression of PDGF-D and PDGFRβ in 58 paired specimens of gastric carcinoma and tissues adjacent carcinoma.Results The positive rates of PDGF-D mRNA and PDGFRβ mRNA(98.28%,87.93%)were higher in gastric cancer tissues than(72.41%,53.45%)in tissues adjacent carcinoma respectively.While the positive rate of PDGFRβmRNA was 72.41%,which was positively correlated with PDGF-D(r=0.40,P0.05).PDGF-D expression was correlated with lymph node metastasis and relapse(P0.05).No correlation was found between PDGF-D and other clinicopathological features of gastric carcinoma(P0.05).Conclusion PDGF-D may play a crucial role in carcinogenesis,development,metastasis and relapse of gastric cancer.It can be used as prognostic markers of gastric cancer in clinical practice.
