摘要
初步构建了菠萝种质的SSR指纹图谱,为菠萝种质鉴定提供了依据。从菠萝EST-SSR开发的20对微卫星引物扩增31份不同的菠萝种质,筛选出能够稳定扩增且带型差异明显的SSR引物。挑选5对多态性高、分辨率高、重复性好的SSR引物对,作为标准引物对材料进行扩增。每对引物得到的多态性带数8~15个,平均为11条;引物的多态信息含量(PIC)在0.643 7~0.792 8,平均0.737 3。依据带型特征,建立菠萝DNA分子指纹图谱。
SSR(simple sequence repeat) markers were used to find reliable fingerprint to identify commercial pineapple cultivars in this study.A total of 31 pineapple accessions were investigated.Five pre-selected primers from 20 EST-SSRs showed ample polymorphism and could distinguish all the materials studied.The mean number of polymorphic bands amplified per primer was 11(range,8 to 15).The mean PIC value was 0.737 3 ranging from 0.643 7 to 0.792 8.The fingerprint code for 31 pineapple accessions were built using 5 EST-SSR primers.
