复方甘草酸苷、地塞米松对毛乳头细胞活性影响的研究 被引量:13

摘要 目的:探讨复方甘草酸苷和地塞米松对体外培养的人毛乳头细胞增殖活性的影响。方法:二步酶消化法分离培养毛乳头细胞,采用免疫组化法对细胞α-平滑肌肌动蛋白进行染色鉴定。噻唑蓝(MTT)法检测不同浓度复方甘草酸苷、地塞米松对人毛乳头细胞增殖的影响。结果:与空白对照组比,复方甘草酸苷在10、20、50mg/L浓度时可明显刺激毛乳头细胞增殖,差异有统计学意义(P<0.05),在0.5~20mg/L范围内刺激作用逐渐增强。与空白对照组比,地塞米松在1、2.5mg/L浓度时可明显刺激毛乳头细胞增殖,差异有统计学意义(P<0.05),在0.5~2.5mg/L范围内刺激作用逐渐增强。复方甘草酸苷组较地塞米松组细胞生长状态好,胞浆更为丰富。结论:复方甘草酸苷和地塞米松在抗炎、免疫调节的同时还可能通过调节毛乳头细胞的活性诱导毛发生长。在治疗脱发性疾病方面,复方甘草酸苷可能比传统的糖皮质激素更有优势。 Objective:To observe the effects of Stroner Neo-MinophagenC (SNMC) and dexameth on the activity of dermal papilla cells (DPCs).Methods:Dermal papilla cells were isolated and cultured from human scalp hair follicles by digestion with dispase and collagenase D.The α-smooth muscle actin (α-SMA) was detected by immunohistochemical staining to identify DPCs.Effects of different concentration of SNMC and dexameth on the proliferation of DPCs were measured by MTT.Results:Human DPCs were isolated by the "two-step enzymatic digestion".The immunohistochemistry result showed that α-SMA was expressed in DPCs.The result of MTT showed that the proliferation of DPCs was significantly promoted by 10,20 and 50 mg/L of SNMC (P 〈 0.05),which was a dose-dependent manner within concentrations of 0.5-20 mg/L.Compared with the control,1 mg/L and 2.5 mg/L of dexameth significantly promoted the proliferation of DPCs (P 〈0.05),which was dose-dependent within concentrations of 0.5-2.5 mg/L.Observed under inverted microscope,the cells of SNMC group had more abundant cytoplasm than those of dexamethasone group.Conclusion:The SNMC and dexameth may induce hair growth by regulating the activity of DPCs in addition to the effect of anti-inflammatory,immune regulation and suppression.The SNMC may have more advantages than conventional corticosteroid in the treatment of hair loss diseases.
