摘要
目的:研究含氮酮吲哚美辛巴布剂、不含氮酮吲哚美辛巴布剂和市售乳膏剂体外透皮吸收特性。方法:以自制无氮酮吲哚美辛巴布剂和市售吲哚美辛乳膏剂作为对照组(n=6),以自制含2%氮酮吲哚美辛巴布剂作为试验组(n=6),采用改良Franz扩散池和小鼠离体皮肤进行体外渗透试验,利用高效液相色谱(HPLC)法对接收液中吲哚美辛浓度进行测定,计算制剂中吲哚美辛累积渗透量和渗透速率。结果:吲哚美辛累计渗透量与时间呈线性关系,为零级动力学特征,3种制剂的渗透速率分别为71.3、47.8和50.7μg·cm-2·h-1。含氮酮吲哚美辛巴布剂较其不含氮酮巴布剂及乳膏剂的体外透皮速率显著增大(q分别为7.6196、6.6780,P<0.01),不含氮酮巴布剂与乳膏剂的体外透皮速率差异无统计学意义(q=0.9416,P>0.05)。结论:含氮酮吲哚美辛巴布剂体外透皮速率较快,具有应用价值。
Objective:To study the transdermal characteristics of indomethacin cataplasm containing azone 、indomethacin cataplasm with no azone and commercially available cream in vitro.Methods: Using rat skin penetration test in vitro with modified Franz diffusion cell,the concentration of indomethacin was determined by HPLC method.The cumulative penetration amount and penetrating rates of indomethacin were calculated.Results: The cumulative penetration amount of indomathacin was linearly related to time ,penetrating rates of these three preparations were 71.3,47.8 and 50.7 μg·cm-2·h^-1 respectively .The penetrating rates of cataplasm containing azone were increased significantly than the cataplasm with no azone or the cream(the value of q were 7.619 6,6.678 0 respectively,P 〈 0.01).There were no significant difference in the penetrating rates between the cataplasm with no azone and the cream(q = 0.941 6,P 〉 0.05).Conclusions:The indomethacin cataplasm containing azone has better penetrating rate,which has applicating value.
