Apak的组织表达谱及其白血病相关性分析

摘要 目的探讨P53负调控蛋白Apak的组织分布特征并分析其白血病相关性。方法利用组织斑点杂交分析Apak在多种组织中的分布情况,Northern杂交分析其在5种白血病细胞系的转录,并用RT-PCR分析Apak在32例白血病患者和12例健康人外周血中的mRNA水平。结果 Apak的组织表达谱分析表明该基因只在胎肝、胎脾以及成人唾液腺、膀胱、左右心室和心尖中高表达。在多种白血病细胞系Jurkat,AHH-1,MOLT-4,SKO-007和HL-60和部分白血病患者的外周血中Apak也呈现高转录,提示Apak在白血病发生中可能发挥重要功能。结论本研究揭示了Apak是一种具有组织特异性分布的P53负调节蛋白,其在白血病中的高表达提示其可能与该病的发生有一定的关联。 Objective To analyze the tissue distribution of Apak and its relationship with leukemia.Methods Dot blot and Northern blot were performed to analyze the tissue distribution of Apak.RT-PCR was used to detect the mRNA level of Apak in five leukemic cell lines.Results Apak was highly expressed in the human fetal liver,spleen and adult salivary gland,bladder,ventricle and vertex cordis.Furthermore,Apak was highly expressed in the peripheral blood of leukemic patients,indicating that Apak might be involved in leukemia.Conclusion Apak is an important tissue-specific regulator of P53 and may be involved in the development of leukemia.
