摘要
本研究建立了抗球虫药乙氧酰胺苯甲酯在鸡组织中残留的紫外HPLC检测方法。经过对鸡各种组织肌肉、肝、肾、脂肪添加高、中、低三个浓度,每个浓度5个样品,共测定2批,证明笔者建立的提取条件的回收率在80% 以上,变异系数在10% 以内。最低标准物检测限为1ng, 最低组织检测限为2.5 ng/g。同时研究了乙氧酰胺苯甲酯在鸡组织中残留的消除规律。试验结果认为以正常剂量乙氧酰胺苯甲酯添加于鸡饲料中,根据颁布的最高残留限量标准,无须休药期,这与美国FDA 规定氨丙啉+
A reversed phase high performance liquid chromatographic method is described for the quantitative residue determination of ethopabate with UV detection in chicken liver, muscle, kidney and fat.The drug is extracted from tissues with acetonitrile, and the extract is concentrated to dry.The residue is dissolved with acetone n hexane(1+9) and cleaned up by solid phase extraction column Florisil, and eluted with methanol.LC analysis is carried out on a Haisil C18 column, and ethopabate is quantitated by using ultraviolet detector set at 270 nm.The drug was eluted with acetonitrile water (30+70) by flow rate of 1ml/min.Average recoveries from chicken tissues fortified with 2MRL, MRL and 1/2MRL of the compound in the individual tissue were ranged from 87.05% to 91.27%, 81.58% to 89.61% , 83.85% to 85.53%, 90.26% to 94.55% from liver, muscle, kidney and fat, respectively.Coefficients of variation were ranged from 6.82% to 10.40%,4.48% to 9.62%,3.94% to 8.76%, 3.33% to 8.07% from liver, muscle, kidney and fat, respectively.The detection limit for ethopabate standard was 1 ng on column, and level as low as 2.5 ng/g in chicken tissues. The residue depletion of ethopabate with amprolium hydrochloride in chicken tissues was studied with 210 AA broiler chicken administered by feed addition of combination of ethopabate and amprolium hydrochloride.The drug was extracted and analyzed by the above developed method.The results showed that there were no residues higher than MRL in each tissue of broiler chicken from both treated groups at the recommended dosage of 8 g/t ethopabate with 125 g/t amprolium hydrochloride and 2 times recommended dosage of 16 g/t ethopabate with 250 g/t amprolium hydrochloride.The results suggested that no withdrawal time was necessary for the application of ethopabate with amprolium hydrochloride.
