摘要 目的:观察不同浓度的葡萄糖对胰岛β细胞脂质含量以及脂肪酸转位酶(CD36)表达的影响。方法:分别以5、10、15、20、253、0 mmol/L葡萄糖孵育胰岛β细胞24 h后,测定细胞内甘油三脂(TG)含量及CD36基因表达。结果:(1)在高糖环境下,细胞内TG含量呈浓度依赖性增加。(2)高糖对CD36的基因表达亦有明显的浓度依赖性。结论:高糖对胰岛β细胞CD36表达的影响可能在脂质异位沉积和脂毒性中有着重要的意义。 Objective:To observe the effects of various levels of glucose on the intracellular triglyceride(TG) content and CD36 expression in β-cells.Methods: NIT-1 cells were incubated with various concentrations of glucose(5,10,15,20,25,30 mmol/L) for 24 h.The content of triglyceride and expression of CD36 mRNA were detected respectively.Results :(1)The TG content in NIT-1 cells increased in dose-dependent manner.(2)Glucose also promotes CD36 expression of NIT-1 cells in a dose-dependent manner.Conclusion:High glucose may play an important role in intracellular triglyceride content and the development of lipotoxicity in pancreatic beta cell.
