RAPD在柑桔珠心苗鉴定上的应用

Discrimination of nucellar seedlings of citrus with RAPD markers

以刘本橙 （刘勤光×本地早） ×代代及其 ３ 个实生苗后代为试材， 用 Ｒ Ａ Ｐ Ｄ 标记对其子代进行鉴定。从 ５０ 个引物中 筛选出 １２ 个引物能在父母本及子代上扩增出清晰的带型， 其中有 １１ 个引物能在父母本间扩增出差异， １ 个引物（ Ｓ２２８） 差异不明显。上述 １２ 个引物中有 １１ 个引物在 ３ 个子代间扩增带型相同， 且与母本带型一致； 另 １ 个引物（ Ｓ２０２） 在母本上的扩增带 （１ ２００ ｂｐ） 在 ２ 个子代中没有出现。３ 个子代中都没有来自父本的特异扩增带。结果表明， ３株实生苗均为珠心苗。

Discrimination of 3 seedlings of citrus Luiben×Daidai with RAPD markers was studied.Of 50 primers screened,12 amplified clear bands in the parents and three seedlings.11 of the 12 primers can produce polymorphism in the male and female parents.Amplification profiles between the 3 seedlings are similar with each other and same as female parent amplified by 11 of the 12 primers.Characteristic bands of male parent didn′t appear in the 3 seedlings.However,a band(1 200 bp)with primer S202 was disappeared in 2 seedlings.It is deduced that the 3 seedlings are from nucellars.
