摘要
以冬虫夏草菌体量为指标,研究了培养基中不同成分对虫草生长的影响,利用Plackett-Burman(PB)试验设计筛选出影响虫草菌生长的关键培养基成分,然后利用最陡爬坡试验和中心组合设计试验优化和确立最佳培养基配方,结果表明,葡萄糖、酵母粉和KH_2PO_4对虫草的生长有显著影响,优化后的培养基成分为葡萄糖59.35g/L、KH_2PO_41.47g/L酵母浸粉16.83g/L、蛋白胨10g/L、MgSO_4·7H_2O0.1g/L。利用此培养基获得的虫草菌体量为26.86g/L,是优化前的2.5倍,为虫草工业化生产提供参考。
Using mycelium biomass as evaluation parameter, effects of medium components on growth of Cordyceps militaris were investigated. The critical components were screened with Plackett-Buman experimental design and the optimum composition of medium was determined with the steepest ascent method and Center Composite Design (CCD). The results showed that glucose, KH2PO4 and yeast extract had the most significant effects on myeelium biomass. The optimum medium for growth of mycelium were as follows, 59.35g/L glucose, 16.83g/L yeast extract, 1.47g/L KH2PO4, 10g/L peptone and 0.1g/L MgSO4.7H2O. By optimization of culture conditions, the Yield of mycelium biomass reached 26.86g/L, which was 2.5 times as that of original medium, when optimum medium was used. It is indicated that application of optimized medium had much potential for improved rnycelium biornass of C. militans in industry.
