发酵液中阿卡波糖的超滤和纳滤膜提取工艺 被引量:4

Process for Extraction of Acarbose from Fermentation Broth by Ultrafiltration and Nanofiltration Membranes

摘要 用超滤和纳滤膜分离技术提取发酵液中的阿卡波糖。发酵液通过超滤膜除去杂质,然后将超滤液通过纳滤膜除盐和浓缩。筛选了不同型号的超滤膜和纳滤膜,并分别优化操作条件,得到适宜的工艺条件:选用ULT-1超滤膜,阿卡波糖发酵液的进料浓度3 g/ml,操作压力0.26~0.28 MPa,操作温度25℃;选用STM-2纳滤膜,操作压力0.6~0.8 MPa,操作温度25℃,料液浓缩倍数为6倍。应用上述条件提取发酵液中的阿卡波糖,提取率为62.2%,较原工艺提高了约20%,浓缩液中阿卡波糖含量高于98.8%。 Ultrafiltration and nanofiltration membrane were used for the separation and extraction of acarbose from fermentation broth.The impurities were removed from the fermentation broth through the ultrafiltration membrane and then the ultrafiltration solution was desalted and concentrated through the nanofiltration membrane.The ultrafiltration and nanofiltration membranes with different molecular weight cutoff were screened and the operation conditions were optimized.Ultrafiltration was performed with ULT-1 membrane and fermentation broth containing and 3 g/ml of acarbose at 25 ℃ and 0.26-0.28 MPa.Nanofiltration was carried out with STM-2 membrane at 25 ℃ and 0.6-0.8 MPa.The final concentration of acarbose was six times higher than the initial one and the extraction yield was 62.2%,a 20% increase compared with the original process.The content of acarbose in the concentrated solution was over 98.8%.
