摘要
目的介绍一种简易高效的大鼠原代肝细胞分离方法。方法在Selgen两步胶原酶灌注法基础上加以改进,进行大鼠原代肝细胞分离。结果平均每只成年SD大鼠获取(1.56±0.31)×108个肝细胞,平均肝细胞活率(95.1±4.2)%,平均肝细胞纯度98%。结论本试验介绍方法简便易行,且肝细胞产量高、活力好、纯度高,适宜在一般条件实验室推广应用。
Objective Introduce a facility and high efficiency separation method of rat primary hepatocytes.Methods Rat hepatocytes were isolated and purified by using the modified Selgen's two-step method.Results The average yield of rat hepatocytes was(1.56±0.31)×108 percent rat,with an average viability of(95.1±4.2)% and purity of 98%.Conclusions The separation method of hepatocytes was simple and easy,with advantages of high yield,good viability and high purity,so it would be applied widely in ordinary laboratory.
