使用Agilent Eclipse XBD-C18分析柱(150mm×6.0mm,5μm),流动相为甲醇(A)∶水(B)=35∶65等度洗脱,之后运行6min,流速1.0ml/min,二极管阵列检测器,检测波长360nm,柱温30℃,构建反相高效液相色谱(reversed phase-high performance liquid chromatography,RP-HPLC)法对丁香中槲皮素进行测定。槲皮素在0.1μg~1.5μg范围内呈良好线性关系,相关系数为0.9991。重复性试验测得槲皮素的平均含量为0.12mg/g,RSD(relative standard deviation)为1.45%。加样回收率为98.9%,RSD为1.16%(n=5)。本方法的优点是准确可靠,分离度好,灵敏度高,重现性好,可用于丁香的质量控制。
An RP-HPLC method was established to determine quercetin for quality standard research of Eugenia caryophllata Thunh. The chromatographic separation was achieved on an Agilent Zorbax SB-C18 (6.0mm × 150mm, 5μm) analytical column using an isocratic elution consisting of methanol (A): H2O (B) 35% : 65% at a flow rate of 1.0 ml/min and 30℃ column temperature, and the analytes were detected using a DAD detector at 360 nm wavelength. The quercetin had fine linear responses within the ranges 0.1 μg - 1.5μg (R^2 =0. 9991 ). The average content of quercetin in the repeatability tests was 0.12mg/g and RSD (relative standard deviation) was 1.45%. The average coefficient of recovery was 98.9% and RSD (n = 5 )was 1.16%. This method is accurate, reliable and has good resolution and repeatability, so it is suitable for quality control of Eugenia caryophllata Thunh.
