几种提取液对苹果成熟叶片总RNA提取质量的影响

Effect of Three Extract Solutions on Total RNA Quality Extracted from Mature Apple Leaves

[目的]筛选一种快速有效地提取高质量的苹果成熟叶片总RNA的提取液。[方法]以红富士苹果成熟叶片为材料,用3种不同的提取液提取苹果成熟叶片的总RNA,并通过分光光度计、琼脂糖凝胶电泳和RT-RCR检测RNA质量。[结果]3种试剂中2种可以提取出总RNA。[结论]用RNAiso-mate for Plant Tissu和RNAiso Plus搭配使用和RNAiso for Polysaccharide-rich Plant Tissue单独使用均可提取出质量较好的RNA,前者质量优于后者。

[Objective]The study was conducted to screen a fast and efficient extract solution for total RNA extraction from mature apple leaves.[Method]Three extract solutions were used for RNA extraction,and RNA was detected by spectrophotometer and agarose gel electrophoresis,and evaluated by RT-PCR.[Result]Two of the three extracts were suitable for total RNA extraction from mature apple leaves.[Conclusion]The RNAiso-mate for Plant Tissue combined with RNAiso Plus,and RNAiso for Polysaccharide-rich Plant Tissue were suitable for high quality RNA extraction from mature apple leaves,but the former solution was better than the latter.
