苯酚-硫酸法测定金顶侧耳中多糖含量 被引量:16

摘要 [目的]建立金顶侧耳多糖含量的测定方法。[方法]通过单因素试验和正交试验确定了苯酚-硫酸法测定金顶侧耳多糖的试验条件。[结果]影响测定结果的主要因素为苯酚用量,其次为浓硫酸用量和显色温度,显色时间影响最小。最佳试验条件为:苯酚0.2ml,浓硫酸5.0 ml,显色温度100℃,显色时间30 min。在最佳试验条件下,葡萄糖标准溶液浓度在20~140μg/ml范围内与吸光度值具有良好的线性关系,线性方程为y=0.004 86x+0.094 60,r=0.999 6;该显色体系在20~120 min内稳定,可用于金顶侧耳多糖含量的测定,测定结果为3.8%,RSD=1.14%(n=6)。[结论]该研究可为金顶侧耳产品中多糖含量的测定提供参考。 [Objective] The aim was to establish the determination method of polysaccharide in P.citrinopileatus.[Method] Determination of polysaccharide in P.citrinopileatus by phenol-sulfuric acid method was studied by two ways of single-factor experiment and orthogonal test.The main factor influencing the determination result was the amount of phenol,the amount of sulfuric acid was next,the water bath temperature was third and the water bath time was the smallest.The optimum experimental conditions were as follow: 0.2 ml phenol,5.0 ml sulfuric acid and 100 ℃water bath for 30 min.In the optimum experimental conditions,the standard solutions of glucose possessed good linear with absorbance when its concentration was 20-140 μg/ml.The linear equation was y=0.004 86x+0.094 6,and the correlation coefficient was 0.999 6.The color apparent system was special stability within 20-120 min.3.8% of mass fraction of polysaccharide extracted from P.citrinopileatus was obtained using the color apparent system and RSD=1.14%(n=6).[Conclusion] The study can provide reference for determination of polysaccharide content from P.citrinopileatus.
