摘要
[目的]对无籽西瓜的组织培养技术进行探索。[方法]以墨童无籽西瓜子叶作为外植体,分别置于添加不同浓度6-BA、NAA、IBA、KT的MS培养基中诱导愈伤组织、丛生芽及丛芽的伸长。[结果]浓度0.3~1.2 mg/L6-BA、NAA均能诱导愈伤组织;浓度0.6~2.5 mg/L6-BA、IBA均能诱导丛生芽;浓度0.2~1.0 mg/LKT均能诱导丛生芽伸长。[结论]诱导无籽西瓜愈伤组织的最佳培养基为MS+NAA1.0 mg/L+6-BA0.5 mg/L,诱导丛生芽的最佳培养基为MS+6-BA2.0 mg/L+IBA0.8 mg/L;诱导丛生芽伸长的最佳培养基为MS+KT0.2 mg/L。
[Objective] The aim was to explore tissue culture technique of Motong seedless watermelon.[Method] Cotyledon of seedless watermelon was made as the explant.The explants were cultured on MS medium with different concentrations of 6-BA,IAA,IBA and KT for inducing callus,clumpy buds and clumpy bud enlongation.[Result] The concentrations from 0.3 mg/L to 1.0 mg/L of IAA,6-BA are able to induce callus;the concentrations from 0.6 to 2.5 mg/L 6-BA,IBA could induce bud;the concentrations from 0.2 to 1.0 mg/L KT could induce shoot elongation.But there were many differences.[Conclusion] The results showed that the best hormone combination of inducing callus was MS+NAA 1.0 mg/L+6-BA 0.5 mg/L.The best hormone combination of inducing clumpy buds was MS+6-BA 2.0 mg/L+IBA 0.8 mg/L.The best hormone combination of clumpy bud enlongation was MS+KT 0.2 mg/L.
