摘要
[目的]检测不同基因型杜仲叶片中的绿原酸含量。[方法]以超声波助提法提取绿原酸,采用HPLC法检测和二倍体含量,色谱柱为Eclipse xdb-c18柱(4.6 mm×150.0 mm,5μm),以乙腈-0.4%磷酸水(10∶90)为流动相,检测波长为327 nm。[结果]四倍体和二倍体杜仲叶片中绿原酸的平均含量分别为4.005和为0.850 mg/g。[结论]四倍体杜仲叶片中绿原酸含量显著高于二倍体。
[Objective]Chlorogenic acid content in eucommia ulmoides of different genotypes were detected.[Method]Ultrasonic was used to extract chorogenic acid.The content of Chlorogenic acid in Eucommia ulmoides at different genotraction was determined by HPLC method.The chromatographic column was xdb-c18(4.6 mm × 150.0 mm,5 μm) and the mobile phase consisted of acetonit rile-0.4% H3PO4 solution(10∶ 90) with a detection of 327 nm wavelength.[Result]The results showed that average content of chlorogenic in tetraploid eucommia ulmoides was 4.005 mg/g,and average of chlogenic in diploid eucommia ulmoides was 0.850 mg/g.[Conclusion]Multiple comparison found that the tetraploid eucommia ulmoides of chlorogenic acid were significantly higher than the diploid content of chlorogenic acid in leaves.
