SX-1液对肝脏保存效果的形态学观察

摘要 目的探讨新研制的肝脏灌注保存液SX-1液对保存肝脏的形态学影响。方法 使用SX-1液、威斯康星大学保存液(UW液)和高渗枸橼酸盐腺嘌呤液(HC-A液)保存大鼠肝脏2h、8h、24h后行大鼠原位肝移植(OLT),于移植后6h处死大鼠提取肝脏组织在光镜进行形态学观察,比较各组保存液对肝脏的保存效果。另外,取SX-1液组保存的肝脏置透射电镜下进行形态学观察。结果 2h组中,3种不同保存液组间比较无明显差别;8h组中,SX-1液组在肝细胞形态学改变的程度和范围上稍比UW液组明显,但明显轻于HC-A液组;24h组中,3组间无明显差别。透射电镜观察SX-1液所保存的肝脏超微结构,显示保存8h范围内肝脏的基本结构形态未见明显毁灭性的改变。结论 在8h保存范围内,SX-1液可以代替UW液和HC-A液。 Objective To observe morphological changes of experimental liver grafts preserved in SX-1 solution.Methods The rat liver grafts were preserved in SX-1 solution,University of Wisconsin(UW)solution and hypertonic citrate adenine(HC-A)solution for 2,8 and 24 hours respectively before orthotopic liver transplantation were performed.Morphological changes of liver grafts were observed when the rats survived for 6 hours after the operation.Results At 2 h and 24 h,there were no significant differences in morphological changes among the three different preservation solutions.At 8 h,the liver injury was more severe in SX-1 solution group compared with the UW solution group,but was alleviated compared with the HC-A solution group.Conclusion The SX-1 solution may replace the UW solution and HC-A solution completely for liver grafts preservation in 8 hours.
