摘要
本文应用选择性抽提整装电镜观察,研究地塞米松诱导人胃癌 M Gc803 细胞分化过程中细胞核骨架中间纤维系统的构型变化。 M Gc803 细胞核骨架纤维和中间纤维数量较少、分布不均匀,核纤层为厚薄不一结构,与两类纤维联系不密切。经 10μg/m l Dex 处理后,细胞核骨架纤维和中间纤维数量增多、结构层次丰富、单丝成份多,分布均匀并相互交织成规则网络,两类纤维通过薄层均一的核纤层发生密切联系,形成贯穿整个细胞核质区域的完整体系。结果表明, Dex 诱导处理细胞的核骨架中间纤维系统产生了与正常细胞相似的恢复性改变,这种变化是癌细胞恶性表型逆转的重要形态和功能表现。
The configurational changes of the nuclear matrix\|intermediate filament (NM\|IF) system in human gastric adenocarcinoma cell line MGc80\|3 after being induced by dexamethasone (Dex) had been studied by selective extraction\|whole mount electron microscopy.It was revealed by electron microscopy that,the NM and IF filaments in MGc80\|3 cells was relatively few and scattered,not well\|distributed and arranged irregularly.The nuclear lamina was nonuniformly thick and compact,connected to NM and IF filaments relaxedly.But in the cells induced by 10μg/ml Dex,the two kinds of filament were plentiful and well\|distributed,differed in slender and thick form and interweaved into a regular network.The NM and IF filaments connected closely by thin and compact fiber\|like lamina,and form a regular network through the whole cell region.It demonstrated that,the NM\|IF system in MGc80\|3 cell had been undergone a restorational change similar to that of normal cells after Dex treatment,and this alternation is an important morphological and functional expression to the maligant phenotypic reversion of cancer cells.
