小麦条锈病不同抗性材料中分子标记YrY201-1分布研究

摘要 以22个抗病品种、4个感病品种以及12个野生近缘材料的基因组DNA为模板进行PCR扩增,检测小麦抗条锈病基因分子标记YrY201-1的分布。结果表明:YrY201-1具有较高的扩增效率(96.2%,25/26);在4个感病材料中均能扩增出目标条带;在野生近缘材料中扩增效率也较高(91.7%,11/12),且在多数材料中扩增条带较多,除目标产物外还有许多非特异性扩增产物出现;在抗性基因YrY201的来源材料J11(粗山羊草)中该标记只扩增出1条特异性条带,但比预期目标条带稍大。 In this study,the DNA from 22 disease-resistant varieties,4 feeling cultivars and 12 wild relatives were used as templates and the distribution of molecular markers YrY201-1 of the stripe rust resistance gene YrY201 was speculated by PCR.The results showed that: the amplification of molecular marker YrY201-1 possessed high efficiency(96.2%,25/26).Target band could be amplified by this marker from all the 4 susceptible materials.The amplification efficiency of the 12 wild relatives was also very high(91.7%,11/12).And several non-specific bands except the target band were amplified from most of the materials.There was only one specific amplification band in the J11(Aegilops) which was the source of molecular markers YrY201-1 for stripe rust resistance gene YrY201,but slightly larger than the prospective band.
