治疗糖尿病中药复方有效部位的研究

摘要 目的探讨中药复方Q治疗糖尿病的主要有效部位及化学成分。方法建立糖尿病小鼠模型。用水提醇沉法将中药复方Q分为20%醇溶部位(20%CR),80%醇沉部位(80%CC)和80%醇溶部位(80%CR)。测定给药前后1 h血糖,血清、肾组织的一氧化氮(NO)和丙二醛(MDA)含量。用薄层色谱法初步分析有效部位的主要成分。结果经分析,80%CC部位主要含有多糖类物质。结论薄层色谱法分析结果证明药味地黄在该复方中起重要作用,结合药效学结果,该方的主要有效成分应为多糖类。 To study the main effective parts and main chemical compositions of Chinese medicinal formula Q for treating diabetes.Methods Formula Q was divided into three fractions by water extracting and alcohol precipitation technology,which were 20 % ethanol-soluble fraction(20 %CR),80 % ethanol precipitation fraction(80 %CC) and 80 % ethanol-soluble fraction(80 %CR).The diabetic mice model was induced by a single i.p.injection of STZ.The blood glucose was assayed at 1 h before and after administration.The contents of NO,MDA in serum and kidney were assayed.The main compositions in the effective parts were preliminarily analyzed by TLC.Results At 1 h after intragastric administration,the blood glucose changed significantly in DM-Q,DM-20 %CR,DM-80 %CC and DM-80 %CR group compared with DM group.There is little variance in the decrease of blood glucose among DM-20 %CR,DM-80 %CC and DM-Q group.But the hypoglycemic activity in DM-80 %CR group was lowered markedly compared with DM-Q group.Compared with DM group,the contents of NO,MDA in serum and kidney were decreased significantly in all the treatment groups.Chemical analysis showed that polysaccharides mainly existed in 80 %CC fraction.Conclusion The activities of inhibiting oxidative stress of 80 %CC and 80 %CR fraction can be close to formula Q.However,the activity of 20 %CR fraction can be stronger.It suggests that 80 %CC and 80 %CR fraction may have a synergistic action in improving oxidative stress.80 %CC fraction has the far more powerful hypoglycemic activity than 80 %CR fraction,even than formula Q and 20 %CR fraction.It can be supposed that the hypoglycemic activity mostly comes from 80 %CC fraction.Based on the pharmacodynamics,the results of TLC analysis further proves that the most important chemical composition in formula Q is polysaccharides from herb rehmannia.
