酸性成纤维细胞生长因子对顺铂所致血管内皮细胞损害保护作用的实验研究 被引量:1

摘要 目的研究酸性成纤维细胞生长因子(aFGF)对顺铂(DDP)引起的体外培养的血管内皮细胞损害的保护作用。方法通过组织块法获取血管内皮细胞、Ⅷ因子相关抗原免疫细胞化学染色鉴定,纯化后传代接种于96孔培养板:(1)培养24 h后加入一系列浓度的DDP,再培养24 h后用MTT法检测细胞存活率;(2)培养24 h后在系列DDP浓度的基础上加入一定浓度的aFGF,继续培养24h后用MTT法检测细胞存活率。结果 aFGF能使DDP对血管内皮细胞的半数抑制浓度(IC50)升高。结论 aFGF对DDP损伤的血管内皮细胞有一定的保护作用。 Aim To investigate the protective effects of acidic fibroblast growth factor(aFGF) on the injury of cultured vascular endothelial cells induced by DDP.Methods The vascular endothelial cells were obtained through tissue mass and identified by immunocytochemistry of Ⅷ factor related antigen.After propagation and purification,the cells were inoculated in 96 well plates:(1)After 24 h cultivation,DDP with different concentrations was added into the cultures,and MTT assay was performed to test the cell viability after 24 h.(2)After 24 h cultivation,aFGF with certain concentrations and DDP with different concentrations were added into the cultures at the same time,and MTT assay was used to test the cell viability.Result The concentrations of DDP that inhibited 50% cell growth(IC50)increased because of the effects of aFGF.Conclusion aFGF can exert certain protective effects on the vascular endothelial cells damaged by DDP.
