葡萄籽原花青素对小鼠深低温脑缺血-再灌注损伤的影响 被引量:2

Effect of grape seed proanthocyanidin extract on deep hypothermic ischemia-reperfusion injury in mice

摘要 目的探讨葡萄籽原花青素(GSPE)对小鼠脑缺血-再灌注(I-R)损伤的保护作用。方法 60只C57/BL6小鼠随机均分为GSPE(A)组、模型(B)组和假手术(C)组,建立深低温I-R模型,再灌注24 h后处死小鼠取脑,RT-PCR技术检测小鼠脑组织半胱天冬氨酸蛋白酶(Caspase)3mRNA,TUNEL法检测小鼠大脑皮层细胞凋亡。结果与C组比较,A组和B组Caspase-3 mRNA表达和细胞凋亡明显增加(P<0.05);而A组各观察指标低于B组(P<0.05)。结论 GSPE对深低温I-R损伤鼠脑有保护作用。 Objective To investigate the protective effect of grape seed proanthocyanidin extract(GSPE) on cerebral ischemia-reperfusion(I-R) injury in mice.Methods Sixty C57BL6 mice were randomly and equally divided into GSPE group(A),model group(B) and sham operated group(C).The deep hypothermic I-R model was established and the mice were sacrificed 24h later for sampling brain tissues.Caspase-3 mRNA was detected by RT-PCR technique and cell apoptosis was analyzed by TUNEL staining.Results Compared with group C,the expression of Caspase-3 mRNA and cell apoptosis in groups of A and B were significantly increased(P0.05),which were lower in group A than those in group B(P0.05).Conclusion GSPE can protect the brain from deep hypothermic I-R injury in mice.
