摘要
目的探讨安宫牛黄丸对大鼠脑出血(intracerebral hemorrhage,ICH)后肿瘤坏死因子(TNF-α)表达的影响。方法将SD大鼠随机分为正常对照组、假手术组、脑出血组、安宫牛黄丸治疗组。采用自体股动脉血注入尾状核建立大鼠脑出血模型,HE染色计数各组48h血肿周围炎性细胞个数,RT-PCR分析各组TNF-αmRNA的表达,酶联免疫吸附测定法(ELISA)检测各组TNF-α蛋白的表达变化。结果安宫牛黄丸治疗明显减少大鼠实验性脑出血后血肿周围炎性细胞个数,抑制脑出血后TNF-αmRNA,TNF-α蛋白的表达。结论安宫牛黄丸能够有效降低大鼠脑出血后TNF-α的表达,对脑出血后的炎症反应具有抑制作用。
Objective To study the effect of Angong Niuhuang Granula on the expression of TNF-α after intracerebral hemorrhage(ICH) in rats.Methods SD rats were randomly divided into normal control group,sham operation group,ICH group and Angong Niuhuang Granula group.The intracerebral hemorrhage model was established by injecting of auto-non-anticoagulant artery blood into the caudate nucleus of the rats.The inflammatory cells around haematoma were counted through HE staining,and RT-PCR assay was adopted to detect the expression of TNF-αmRNA,ELISA assay was used to determined the expression of TNF-αprotein.Results Angong Niuhuang Granula could significantly alleviate the number of inflammatory cells around haematoma,inhibit the expression of TNF-α mRNA and protein.Conclusion Angong Niuhuang Granula can reduce the TNF-αmRNA and protein expression,inhibit the inflammatory reaction after ICH in rats.
