用ELISA检测传染性囊病病毒诱导的细胞凋亡

Detection of Apoptosis of Chicken Embryo Fibroblasts Induced by Infectious Bursal Disease Virus Using ELISA

用5株传染性囊病病毒（IBDV）分别感染鸡胚成纤维细胞（CEF），于感染后不同时间收集接毒组与对照组的细胞进行检测．试验结果显示，接毒组与对照组细胞的DNA在琼脂糖凝胶电泳谱上均呈现梯状条带．用酶联免疫吸附试验（EUSA）检测各组细胞中降解DNA量，发现感染IBDV7h后，接毒组细胞降解DNA量比对照组明显增加，且各接毒组间降解DNA量有一定的差异．试验结果表明：各株IBDV均诱导CEF发生细胞凋亡，但不同IBDV毒株诱导细胞凋亡的能力有差异；ELISA是定量检测细胞凋亡的较好方法．

Chicken embryo fibroblasts (CEF) infected with each of five stains of infectious bursal disease virus(IBDV) and control CEF were harvested and tested at various stages post-infection. Upon Agarose gel electrophoresis, DNA extracted from IBDV-infected and control CEFs exhibited a laddering Pattern. The amount of the DNA fragments was measared with enzyme-linked immunosorbent assay (ELISA). There was a significant increase in the amount of DNA fragments in each IBDV-infected group, as compared with the control group, and the amount of DNA fragmets showed some differences among the infected groups. These results demonstrated that the five IBDV strains all could induce apoptosis, but different IBDV strains exhibited variation in the capacity to induce apoptosis; quantitative change in apoptosis was better detected with ELISA.
