摘要
目的建立HPLC法测定拟青霉中活性成分腺苷含量的分析方法。方法色谱柱为kromasil-C18(150 mm×4.6 mm,5μm),流动相为乙腈:水=5:95,流速为1.0 ml/min,检测波长为260 nm,柱温为室温。结果腺苷的线性方程为Y=0.476 4X-0.376 6,线性范围2.56~12.8μg/mlr,=0.999 6,平均回收率为99.51%,RSD为1.44%。结论该方法灵敏度高、选择性好,样品前处理方法简单快速,回收率符合实验要求,适合于拟青霉中活性成分腺苷的含量分析。
Objective To establish a determination method for adenosine in paecilomyces by HPLC.Methods A kromasil C18(150 mm×4.6 mm,5 μm) column was used.The mobile phase consisted of acetonitrile and water(5:95) at a flow rate of 1.0 ml/min.The detective wavelength was at 260 nm.Column temperature was room temperature.Results Adenosine linear equation was Y=0.476 4X-0.376 6 with a linear range of 2.56~12.8 μg/ml,r=0.999 6.The average recovery was 99.51% with RSD of 1.44%.Conclusion The method is sensitive and selective,its pretreatment is simple and fast,and the recovery accords with the requirement of experiment,and it can be used to determine the content of adenosine in paecilomyces.
