间歇运动对心肌肾上腺髓质素特异性受体RAMP_2及CRLR的影响 被引量:3

摘要 目的研究间歇运动(Intermittent exercise)对心肌肾上腺髓质素(Adrenomedullin,ADM)受体活性修饰蛋白2(Receptor activity modifying protein,RAMP2)和降钙素受体样受体(calcitonin receptor like receptor,CRLR)(两者组成肾上腺髓质素特异性受体)的影响,探讨ADM在间歇运动诱导心肌细胞保护中的作用机制。方法 SD大鼠30只,分为对照组(C组),间歇运动组(IE组),IE组进行3 w的间歇跑台运动,应用免疫印记法检测心肌肾上腺髓质素特异性受体RAMP2和CRLR的定量。结果 IE组心肌RAMP2和CRLR定量均明显上升。结论间歇运动使心肌ADM特异性受体CRLR和RAMP2在蛋白水平上调,ADM对于心肌的作用会因CRLR和RAMP2的上调而加强。间歇运动引起的ADM特异性受体的正向调节作用是适度的反应性增强调节,能增强心脏对运动的适应,对在运动中维持血液动力学稳定和保护心肌细胞起一定的作用。 Objective To probe the effect of Intermittent exercise on ADM, Receptor activity modifying protein ( RAMP2 ) and Calcitonin receptor like receptor(CRLR) in myocardial cytoprotection of rat, we study the function and mechinsm of Adrenomedullin (ADM) in myocardial cytoprotection inducd by IE in heart of rat. Method Sprague - Dawley rats( n = 30) were randomly into two groups: control group( C), intermittent exercise group (IE). The rats of IE group performed 3 weeks intermittent treadmill exercise in order to establish animal model. The contents of ADM in serum and myocardium were investigated using euzymelinked immunosorbent assay and RAMP2 and CRLR in myocardium were investigated using Western blot. Result As compared with the control, the content of serum ADM were not obvious change but contents of ADM in myocardium were obvious increased and RAMP2 and CRLR also were obviously increased. Conclusion The results suggested that secretion and releasing of ADM in myocardium by intermittent exercise and its specificity receptor RAMP2 and CRLR also were obviously increased follow in myocardial. That increase of Adrenomedullin and specificity receptor leads to upregulation and exercise adaptit of heart and plays an important role in keeping haemodynamics stabilization and myocardial eytoprotection in exercise.
