摘要
目的: 探讨川芎嗪(TMP)与母药川芎注射液之间的药效关系. 方法: 以8 m g·kg- 1 的磷酸TMP(TMPP)注射液和川芎注射液(0.1 g·kg- 1 )单独和混合,从健康麻醉开胸犬的心房内注射;并测定后者血清中TMP浓度,换算后以低剂量0.1 m g·kg- 1 TMPP注射. 结果: TMPP引起明显的HR增快, LVSP, LVdP/dtm ax 和CBF增加和m AoP下降(P<0.05~0.01), 12 m in~20 m in 内恢复正常;川芎注射液在20m in~120 m in 内,引起非常显著的HR减慢,LVS, PLVdP/dtm ax, CBF和m AoP的降低(P< 0.01),同样剂量混合给药后,两药从药效性质、强度、作用时间和其持续时间上,诱导出和单独给药时一致的、完全相反和不同的效应,后者的血清TMP浓度远低于阈值(7.3±1.4) ng·L- 1),低剂量TMPP无药效变化. 结论: TMP可能与川芎注射液的急性血流动力学效应无关.
AIM: To make a hemodynamic comparison between tetramethylpyrazine (TMP) and Ligusticum chuanxiong Hort (LCH). METHODS: Heart rate (HR), left ventricular systolic pressure (LVSP), peak derivative of left ventricular systolic pressure (LVdP/dt max ), coronary blood flow (CBF) and mean aortic pressure (mAoP) were compared after bolus atrial injection of high (8 mg·kg -1 ) and low (0.1 mg·kg -1 , equal to TMP content in LCH) dose of TMP phosphate (TMPP), and LCH (0.1 g·kg -1 ) injection to open chest anesthetized dogs respectively. Serum TMP concentration was determined after the bolus atrial injection of LCH. RESULTS: TMPP increased HR, LVSP, LVdP/dt, CBF and mAoP, while LCH injection decreased the above parameters ( P <0.01). Serum TMP concentrations of LCH injection were much lower than the minimal effective concentration (7.3±1.4) μg·mL -1 of TMP in serum. When TMPP and LCH were given simultaneously, their pharmacodynamic property, effective time were significantly different or opposite, which were similar to their effects described above. But low dose of TMPP did not have any hemodynamic effect. CONCLUSION: It is possible that TMP is not related with acute hemodynamics of LCH injection.
