摘要
目的: 测定精子体外培养的顶体反应百分率,并建立一套体外培养精子的方法. 方法: 采用普通孵箱培养蟋蟀精子,在光镜水平上测其存活时间,并用凝集素作为分子探针,测其顶体反应百分率. 结果: 精子在Tc-100培养基内存活时间最长,为5.0 h;在PBS和昆虫生理盐水中存活时间最短,为1.5 h;在平衡盐溶液中可存活2.0 h. 咖啡因可明显促进精子的顶体反应百分率. 结论: 体外培养精子是一种简单、快速研究昆虫精子获能和顶体反应的好方法.
AIM: To provide a new method to determine the rate of acromosal reaction in vitro . METHODS: The life time of cricket sperm cultured in incubator was measured under microscope. Lectin was used as a probe to determine the rate of acromosal reaction. RESULTS: The sperm surviving time in Tc-100 medium was about 5.0 h, the longest time, in physiological saline and PBS was about 1.5 h, the shortest time and in balanced saline 2.0 h. The fact that caffeine can obviously enhance the rate of acrosomal reaction was proved in this paper. CONCLUSION: Culturing the sperm in vitro is a more simple and rapid way to study the insect sperm capaciation and acromosal reaction.
