摘要
目的 选择抗生素和化学诱变剂处理雷帕霉素产生菌的原生质体 ,筛选高产菌株。 方法 分别用庆大霉素、丝裂霉素 C、红霉素、亚硝基胍处理吸水链霉菌 FC90 4( S.hygroscopicus FC90 4)原生质体 ,比较它们对雷帕霉素产量的影响 ,并筛选高产菌株。 结果 庆大霉素的处理效果较好 ,从庆大霉素处理株中筛选到高产菌株 C14、C14 - 1,其雷帕霉素的摇瓶发酵水平 (反相高效液相色谱法测定 )分别比原始株提高 36% ,60 %。 结论 用庆大霉素处理吸水链霉菌 FC90 4原生质体是提高雷帕霉素产量的有效手段。
To screen higher producing strains by the treatment of protoplasts of S hygroscopicus FC904 with antibiotics and nitrous guanidine(NTG) \ Methods\ Protoplasts of S hygroscopicus FC904 were treated with gentamicin,erythromycin,mitomycin C or NTG respectively \ Effects of these agents on rapamycin production were compared and the high yielding strains were screened \ Results\ The treatment with gentamicin had a better effect on improving the production of rapamycin \ Higher producing strains C14 and C14 1 were obtained from the gentamicin treated strains with their RPM titer of broth(assayed by HPLC) increased by 36% and 60% respectively as compared with that of the original strain S hygroscopicus FC904 \ Conclusion\ The treatment of S hygroscopicus FC904 protoplasts with gentamicin is effective for improving the production of rapamycin \;
