摘要
目的:研究延边地区朝鲜族糖尿病肾病(diabetic nephropathy,DN)患者NADPH氧化酶p22phox亚基242位点基因多态性分布,探讨延边地区朝鲜族DN患者该基因多态性与DN发病相关情况。方法:应用聚合酶链反应-限制性片段长度多态性技术对延边地区人群中186例朝鲜族DN患者及139名朝鲜族正常对照组者NADPH氧化酶p22phox亚基242位点进行基因型分析,并将住院治疗的186例朝鲜族DN患者分为男性和女性两组,分析两者基因型频率、等位基因频率的差异。结果:①NADPH氧化酶p22phox亚基C242T基因多态性在朝鲜族正常对照组和朝鲜族DN患者中的分布差异无统计学意义(χ2=1.854,P>0.05);②在男性和女性DN的患者中,NADPH氧化酶p22phox亚基242位点纯合子CC基因频率与杂合子CT及纯合子TT基因频率相比,差异有统计学意义(χ2=5.278,P<0.05);结论:①NADPH氧化酶p22phox亚基C242T基因多态性与延边朝鲜族DN患者的发病无关;②CC纯合子基因可能是男性朝鲜族DN患者的易感基因。
Objective To investigate the distribution of NADPH oxidase subunit p22phox gene C242T polymorphism in Korean diabetic nephropathy(DN) patients of Yanbian area,and analyze the association of NADPH oxidase subunit p22phox gene polymorphism with Korean DN patients in Yanbian area.Method Genotypes of p22phox gene were typed in 186 Korean DN patients and 139 Korean normal controls by polymerase chain reaction-restriction fragment length polymorphism;the association between p22phox gene C242T polymorphism with Korean DN patients in Yanbian area was analyzed.Results ①The distribution of NADPH oxidase subunit p22phox gene C242T polymorphism showed no significant difference between normal control group and DN group(χ2=1.854,P0.05);②The distribution of CC homozygous genotype vs CT heterozygous and TT homozygous genotype showed statistically significant difference in p22phox gene C242T between male and female Korean DN patients(χ2=5.278,P0.05).Conclusion ①The distribution of NADPH oxidase subunit p22phox gene C242T polymorphism has no relation with DN;②The CC genotype of the C242T polymorphism of p22phox gene is a susceptibility gene for the male Korean DN patients.
