目的:检测 bcl-2和 Bax在大鼠全脑缺血再灌后海马区的表达。方法:用原位杂交组织化学检测 bcl-2 mR-NA,用免疫组织化学检测Bax蛋白。结果:bcl-2主要在海马CA3区表达,再灌8小时出现,24小时阳性信号达到高峰,72小时明显下降;Bax则主要在CA1区表达,再灌8小时出现,24小时阳性信号达到高峰,72小时明显下降,CA2、CA4、DG区也有表达,但变化不明显。高倍镜下可见紫蓝色的阳性信号和棕黄色阳性信号位于胞浆和突起中,细胞核处淡染。结论:大鼠全脑缺血再灌后海马区 Bax和 bcl-2的区域性表达与缺血性损伤有一定的关系,可能参与海马不同区域缺血耐受性差异的形成。
To study the expression of apoptosis-related gene, bcl-2 and Bax in hippocampus after global ischemia, immunohistochemistry staining and in situ hybridization were used to detect Bax and bcl-2 mRNA respectively. The results showed that both bcl-2 mRNA positive signals and Bax- positive neurons occured at 8-reperfusion-hour, reached max at 24 hours, and reduced markedly at 72 hours after ischemia. BCL-2 mRNA positive signals however, mostly were located in CA3 zone, and Bax-positive neurons in CA1 subfield. Positive signals were located in the cytoplasma. Our findings suggest that selected expression of different members of Bax and bcl-2 gene families may play an important role in determining the relative sensitivity of neuronal subpopulations to ischemia.
