摘要
为探讨Mg^(2+)ATP酶在精子轴丝内的分布而进行此实验。人精子先以TritonX-100脱膜,再进行Mg^(2+)ATP酶的硝酸铅法细胞化学染色。轴丝ATP酶阳性产物以中央微管为中心,呈现为呈同心圆排列的3个环带状分布。从外向内分别与轴丝的外 Dynein臂、内 Dynein臂和放射辐头的结构区带相对应。反应以外带最强,中带与内带相似。研究证明了以往不受人们重视的放射辐头部也有与内Dnnein臂相似的ATP酶的活性。
This experiment was carried out to investigate the activity of Mg^(2+)-ATPase in axoneme of human spermatozoa. After being demembraned by Triton X-100, the spermatozoa were stained by cytochemistry with reaction of lead nitrate for Mg^(2+)-ATPase. The post reactive productions of ATPase were located in the axoneme and formed three concentric circles to surround the two cen- tral microtubules. From inner to outer, the areas which the three circles were deposited at, just corresponded with the positions of radial spork heads, inner dynein arms and outer dynein arms respectively. The intensity of ATPase was greatest at the outer icrcle. The intensity of ATPase reaction at the medium circle was the same as that of ATPase reaction at the inner circle. Our ob- servation demonstrate that the radial spork heads, which once have been neglected by the people, have the same intensity of Mg^(2+)-ATPase activity as inner dynein arms.
