摘要
目的:建立金樱子中熊果酸和齐墩果酸含量测定方法。方法:采用液质联用(LC-MS/MS)法,色谱柱Diamonsil C18(2)(250 mm×2.1 mm,3μm),流动相甲醇-甲酸水(87∶13),流速0.25 ml.min-1。柱温30℃,质谱采用大气压电喷雾离子源,选择性离子流模式(负离子)。结果:熊果酸和齐墩果酸的平均样回收率分别为为98.1%和97.6%,RSD分别为2.7%和2.9%。结论:该方法快速简便,灵敏度高,重现性好。可用于金樱子的质量控制。
Objective:To establish an HPLC-MS/MS method for determination of the content of Ursolic Acid and Oleanolic Acid in Rosae Laevigata michx.Methods:Chromatograthy was performed on a 3μm Diamonsil C18(2) column with the column temperature at 30℃.and the sample size at 10μl.The mobile phase composition was methanol~water(0.2%formic acid)(87∶13).The flow rate was 0.25 ml ·min.Results:The Ursolic Acid had a good linearity relationship(r=0.999 1,n =6) between 5.08~1016 ng/ml;The Oleanolic Acid had a good linearity relationship(r=0.999 2,n=6) between 6.46~1028 ng/ml.The averagerecovery of Ursolic Acid and Oleanolic Acid in Rosae Laevigata michx was 98.1%(RSD:2.7%) and 97.6%(RSD:2.9%).CONCLUSIONS:This method is simple,accurate and reproducible.
