摘要
目的:探讨白藜芦醇对胃癌BGC-823细胞中凋亡相关基因人程序化死亡分子5(PDCD5)表达的影响,并观察白藜芦醇诱导BGC-823细胞凋亡的效应。方法:分别用25、50、100和200μmol/L的白藜芦醇处理BGC-823细胞24、48和72h后,采用免疫细胞化学和原位杂交方法检测PDCD5表达的变化;运用流式细胞仪和TUNEL法检测细胞凋亡情况。另设空白对照组和溶剂对照组。结果:同一时间点,6组PDCD5蛋白及mRNA的表达差异均有统计学意义(F蛋白=211.881、242.438和184.866,P均<0.001;FmRNA=207.366、148.411和158.661,P均<0.001);同一浓度,24、48和72h时点各组比较,PDCD5蛋白及mRNA的表达差异均无统计学意义(F蛋白=0.950、1.870、2.218和2.232,P均>0.05;FmRNA=1.267、2.376、1.221和2.292,P均>0.05)。各组BGC-823细胞凋亡指数比较,差异有统计学意义(F=230.069,P<0.001)。结论:凋亡相关基因PDCD5可能在白藜芦醇诱导胃癌BGC-823细胞凋亡过程中发挥重要作用。
Aim:To explore effects of resveratrol on expression of apoptosis-related gene PDCD5 and apoptosis in gastric cancer cell line BGC-823.Methods:BGC-823 was cultured and treated with different concentrations of resveratrol(25, 50, 100, and 200 μmol/L) for 24,48,and 72 h,respectively.The expressions of PDCD5 protein and mRNA were detected by immunocytochemistry and in situ hybridization, and cell apoptosis was analyzed by flow cytometry and TUNEL.Results:There were significant differences in PDCD5 protein and mRNA expression levels among the 6 groups at 24,48, and 72 h points(Fprotein=211.881,242.438,184.866, P0.001; FmRNA=207.366,148.411,158.661,P0.001).In the same concentration(25,50,100,and 200 μmol/L) of resveratrol group, with the increasing of time,the expression levels of PDCD5 protein and mRNA had no significant difference(Fprotein=0.950,1.870,2.218,2.232,P0.05; FmRNA=1.267,2.376,1.221,2.292,P0.05).BGC-823 cell apoptosis index had significant difference among the 6 groups(F=230.069,P0.001).Conclusion: PDCD5 plays a key role in the gastric cancer cell apoptosis induced by resveratrol.
