摘要 目的:评价PCR短串联重复序列(PCR-STR)在异基因外周血干细胞移植(Allo-PBSCT)后监测中的应用价值。方法:采集13例造血干细胞移植(HSCT)患者首次术后10、20和30d的口腔颊黏膜上皮细胞及外周静脉血,提取基因组DNA,采用PCR-STR法比较其外周血与口腔黏膜细胞中D7S3048、D4S2639、D14S306、D18S535、F13A01、FESFPS、vWA、CSF1PO、TPOX和THO1等STR位点联合基因型改变。结果:13例HSCT患者的外周血中均发现外源STR基因型。10、20和30d13例患者口腔黏膜细胞与外周血检测的10个STR基因位点均至少有5个不同。结论:对受者外周血与口腔黏膜多个STR基因位点的基因型进行对比,可随时监测HSCT患者植入效果。 Aim:To evaluate the value of PCR-STR which applied in monitoring hematopoietic stem cell transplantation(HSCT).Methods:The peripheral blood and epithelial cell of oral mucosa were collected from 13 patients at 10, 20,and 30 days after the first transplantation. Ten STR loci had been amplified by PCR after the DNA extraction.Results: Exogenous gene was found in the peripheral blood of all the patients.There were at least five STR loci different between the peripheral blood and epithelial cell of mucosa in all of 13 patients.Conclusion:Comparison the DNA genotypes between peripheral blood and oral mucosa could be used to monitor the effect of HSCT.
