摘要
目的:检测大、小鼠空肠、肝及肾组织中P型铜转运ATP酶(ATP7B)蛋白的表达。方法:分别取6只SD大鼠和7只昆明小鼠,取空肠、肝及肾组织,常规石蜡切片,采用免疫组化法检测ATP7B蛋白的表达。结果:大、小鼠空肠绒毛上皮的纹状缘和近基底部、小肠腺、肾小管和肝细胞中均有不同程度的ATP7B蛋白表达。大鼠空肠、肝及肾组织ATP7B的积分光密度分别为(224474.380±228536.513)、(94116.484±45821.113)和(118128.741±41046.629),小鼠上述3种组织中ATP7B的积分光密度分别为(129023.602±87570.591)、(150614.511±52981.792)和(118403.634±53656.681),大、小鼠同一组织器官ATP7B的积分光密度相比,差异均无统计学意义(t=1.239、1.355、0.009,P均>0.05)。结论:ATP7B蛋白在大、小鼠空肠、肝及肾组织中均有表达,其表达量的高低与动物种属关系不大。
Aim:To detect the expression of ATP7B in jejunum, liver and kidney tissue of rats and mice.Methods:The jejunum, liver and kidney tissue was excised from 6 SD rats and 7 Kunming mice. The expression of ATP7B was detected using immunohistochemical staining.Results:ATP7B positive immunohistochemical staining was localized in striated border,basilar part of epithelial cells and small intestine glands of jejunum, in the renal tubules for the kidney tissue,and in hepatic cells for the liver tissue,however, the expression levels were different. The integral optical density of ATP7B in rat jejunum, liver and kidney tissue was (224 474.380±228 536.513),(94 116.484±45 821.113),(118 128.741±41 046.629),which in the above tissue of mice was (129 023.602±87 570.591),(150 614.511±52 981.792),(118 403.634±53 656.681),and there was no significant difference in the expression levels of ATP7B between the same tissue of rats and mice(t=1.239,1.355,and 0.009,P0.05).Conclusion: The expression of ATP7B is found in jejunum, liver and kidney tissue of rat and mouse, but the level of ATP7B expression is not related with the animal genus.
