摘要 目的克隆斑马鱼COX-1、COX-2a基因,研究其在斑马鱼胚胎早期发育中的表达情况。方法提取24 h斑马鱼胚胎的总RNA,制备DIG标记的COX-1、COX-2a RNA反义探针,整胚原位杂交研究COX-1、COX-2a在斑马鱼胚胎早期发育过程中的表达。结果 COX-1在囊胚期和原肠胚期为弥散性表达,18 h集中表达在后部躯干,到24 h,COX-1在输尿管和远端脊索处有表达,24~36 h,头部表达量高,48 hCOX-1的表达弱。COX-2a在原肠胚期前不表达,在10.8 h开始表达,位于前端神经外胚层,在26 h,COX-2a在前脑、小脑和后脑有特异性表达。结论明确了COX-1、COX-2a基因在斑马鱼胚胎发育过程中的表达模式,为研究该基因功能提供了一定的理论基础。 Objective Clone cyclooxygenases(COX) gene and investigate expression patterns of COX-1,COX-2a during early embryonic development in zebrafish.Methods Total RNA of 24 hour-stage zebrafish embryos was extracted and COX1,COX-2a digoxin-labeled RNA probe were prepared to investigate the expression patterns of COX-1 and COX-2a during zebrafish embryogenesis by whole mount in situ hybridization of zebra embryo.Results COX-1 was expressed diffusely in blastula stage and gastrula stage.During segmentation stage,intense expression of COX-1 was observed in developing posterior body at 18 h,and eventually became confined to the distal end of the nephric duct,forming vasculature,hypochord,and posterior end of notochord at 24 h.Also,it was expressed in head highly at 24 h and 36 h,it could not be detected almost at 48 h.On the other hand,COX-2a transcript was not detected until the end of gastrulation specifically in the anterior neuroectoderm,it was expressed in hindbrain(cerebellum) and rhombencephalon at 26 h.Conclusion The expression pattern of COX-1,COX-2a during zebrafish embryogenesis was made clear,providing some basis for the function of COX
