摘要
目的观察烟酰胺(NAM)对人脂肪组织来源间质干细胞(hATSCs)增殖的影响。方法将原代培养的hATSCs分别暴露于不同浓度(0、0.05、0.1、0.5、1、5、10、25、50、100μmol/L)的NAM并分别培养24、48、72 h,以MTT方法检测NAM对hATSCs增殖的影响。结果≤25μmol/L的NAM培养244、8、72 h后,对hATSCs的增殖无影响(P>0.05);当NAM浓度≥50μmol/L时,对hATSCs的增殖有抑制作用(P<0.05或0.01)。结论当NAM浓度范围在0~25μmol/L时,对人脂肪来源的间质干细胞的增殖无显著影响;当NAM的浓度≥50μmol/L时,出现细胞毒性作用。
Objective To observe the influence of nicotinamide(NAM) on the proliferation of human adipose tissue-derived stromal cells(hATSCs).Methods Human adipose tissue-derived stromal cells(hATSCs) were cultured primarily.The hATSCs were treated with nicotinamide at the doses of 0.05,0.1,0.5,1,5,10,25,50 and 100 μmol/L for 24 h,48 h and 72 h respectively,and the influence of nicotinamide on the proliferation of human adipose tissue-derived stromal cells(hATSCs) was observed by MTT assay.Results There was no significant influence on the proliferation on hATSCs,when the concentration of NAM was no more than 25 μmol/L whether cultured for 24 h,48 h or 72 h(P0.05);whereas the proliferation of hATSCs was suppressed significantly when the concentration of NAM was over 50 μmol/L whether cultured for 24 h,48 h or 72 h(P0.01).Conclusion With the concentration ranging in 0-25 μmol/L,nicotinamide has no remarkable influence on the proliferation of hATSCs,and when the concentration of NAM is over 50 μmol/L,it has suppression on the proliferation of hATSCs.
