摘要
目的探讨甲基化CpG结合域蛋白1(MBD1)和VIMENTIN蛋白在胰腺癌组织中的表达情况及其与胰腺癌临床病理特征的关系。方法采用免疫组织化学方法检测40例经手术切除的胰腺癌组织和10例良性肿瘤患者的正常胰腺组织中MBD1和VIMENTIN蛋白的表达。结果在40例胰腺癌标本中有28例(70.0%)MBD1呈阳性表达,16例(40.0%)VIMENTIN蛋白呈阳性表达,显著高于正常胰腺组织标本的1例(1/10)和0例(P值均<0.05)。MBD1与VIMENTIN蛋白的阳性表达呈正相关(r=0.423,P=0.012)。在有淋巴结转移的胰腺癌中的MBD1表达阳性率显著高于无淋巴结转移的胰腺癌(P=0.023)。在病理分化较差和有淋巴结转移的胰腺癌中的VIMENTIN表达阳性率显著高于病理分化较好及无淋巴结转移的胰腺癌(P值分别=0.027、0.003)。结论胰腺癌组织中存在MBD1和VIMENTIN的过表达,并与病理分级和淋巴结转移有关,MBD1可能通过调控甲基化过程,干预VIMENTIN的表达,在胰腺癌的发生、发展中起到重要作用。
Objective To investigate the expressions of methyl-CpG binding domain protein 1(MBD1), VIMENTIN and their correlation with clinical pathological parameters in human pancreatic carcinoma.Methods Immunohistochemistry was used to detect the expression of MBD1 and VIMENTIN in 40 pancreatic cancer tissues and 10 normal pancreatic tissues.Results The positive rates of MBD1 and VIMENTIN in pancreatic carcinoma tissues were 28/40(70.0%) and 16/40(40.0%),respectively,which were significantly higher than those in the normal control group(1/10 and 0,both P0.05).The expression of VIMENTIN was significantly correlated with the pathological degrees(P = 0.027) and lymph node metastasis(P = 0.003) of pancreatic cancer,and the expression of MBD1 was correlated with lymph node metastasis(P = 0.023).We also found a positive correlation between MBD1 and VIMENTIN expression in pancreatic cancer(r = 0.423,P = 0.012).Conclusion MBD1 and VIMENTIN are over-expressed in human pancreatic carcinoma,which is correlated with the pathological degree and lymph node metastasis of pancreatic carcinoma.The expression of MBD1 is closely related to the metastasis of pancreatic cancer and expression of VIMENTIN.MBD1 may be involved in regulating VIMENTIN expression by mediating DNA methylation in pancreatic cancer,thus playing an important role in the carcinogenesis of the pancreatic carcinoma.
