摘要
由于牛血清白蛋白(BSA)与光色素的结合反应,引起光色素荧光淬灭,且其荧光淬灭程度与BSA的浓度在0.050~1.0 mg.L-1范围内呈线性关系。据此提出了以光色素为探针测定抗体蛋白质的荧光方法。方法的检出限(3S/N)为8.0μg.L-1。用标准加入法对方法的回收率作了试验,测得回收率在95.0%~104.0%之间,测定值的相对标准偏差(n=6)小于2.5%。
Based on the fluorescence quenching of lumichrome due to its combination with bovine serum albumin(BSA) and the linear relationship between magnitude of intensity of fluorescence quenching of lumichrome and concentration of BSA in the range of 0.050 to 1.0 mg·L-1,a fluorospectrophotometric method for determination of BSA using lumichrome as fluorescence probe was proposed.Detection limit(3S/N) of the method was found to be 8.0 μg·L-1.Recovery and precision of the method were tested by the standard addition method,giving values of recovery in the range of 95.0%-104.0%,and RSD′s(n=6) less than 2.5%.
