摘要
为了优化白灵菇深层发酵的条件,以菌丝生物量为指标,对碳源、氮源、初始pH、接种量、装液量进行单因素试验,结果表明160 r.min-125℃培养条件下,白灵菇最佳液体培养基配方为玉米淀粉4%、米糠5%、KH2PO40.3%、MgSO40.1%、VB110 mg.L-1,最佳装液量为70 mL(250 mL三角瓶),最佳接种量为20%,初始pH为7~8。正交试验结果表明,影响白灵菇液体培养的主次因素依次为接种量、碳源、氮源、初始pH和装液量。
The paper studied the optimization of submerged culture conditions of Pleurotus nebrodensis using the mycelia biomass as the test index.Monofactorial test revealed that mycelial biomass of Pleurotus nebrodensis grown in submerged culture was maximal under the following conditions:160 r·min^-1 temperature 25 ℃,corn starch 4%,rice bran 5%,KH2PO4 0.3%,MgSO4 0.1%,VB1 10 mg·L^-1,the best broth content was 70 mL(250 mL Erlenmeyer flask),the best inoculated amount was 20%,the initial pH was 7~8.Application of orthogonal experiment revealed that mycelial biomass were influenced significantly by inoculated amount,followed by carbon source,nitrogen source,initial pH,broth content.
