麻疹疫苗株和野毒株病毒鉴别方法应用于接种麻疹减毒活疫苗后的麻疹样病例标本鉴定 被引量:11

The Application of Identification Method of Measles Vaccine Strain and Wild Strain on Identifying Specimen from Measles-like Case after Vaccination of Measles Attenuated Live Vaccine

摘要 目的对接种麻疹减毒活疫苗(Measles Attenuated Live Vaccine,MV)后第9d出现的一例麻疹样病例标本,进行疫苗株和野毒株麻疹病毒鉴定。方法采用两种逆转录-聚合酶链反应-限制性片段长度多态性分析(Reverse Transcription-Polymerase Chain Reaction-Restriction Fragment Length Polymorphism,RT-PCR-RFLP)方法,检测麻疹样病例咽拭子标本。结果应用H1基因型麻疹野病毒RT-PCR-RFLP方法,对该病例咽拭子标本进行鉴定,其RT-PCR阳性产物经SalI酶切后,被切成两个片段,分别为408个和159个核苷酸,显示该病例是由H1基因型麻疹野病毒引起的。应用中国MV株RT-PCR-RFLP方法对该病例咽拭子标本进行鉴定,其RT-PCR产物不能被AflⅡ酶切,仍呈单一条带,显示该病例是由非MV株病毒引起的。结论接种MV第9d后出现的麻疹样病例,被证实为H1基因型麻疹野病毒引起,而非MV株病毒引起。 Objective To distinguish measles vaccine strain and wild strain from a measles case after measles vaccination at the 9th day.Method To apply 2 kinds of methods of Reverse TranscriptionPolymerase Chain Reaction-Restriction Fragment Length Polymorphism(RT-PCR-RFLP) to detect NPS specimen of measles.Results The RT-PCR-RFLP method for H1 genotyping was used for identifying this case,and its RT-PCR products were cut into 2 fragments of 408 bp and 159 bp,the results showed that this case was caused by H1 genotype.The RT-PCR-RFLP method for vaccine strain identification was used for identifying this case,and its RT-PCR products couldn't be cut by AflⅡ enzyme and showed a single band.The results showed that this case was caused by non-China vaccine strain.Conclusions The case who got measles after 9 days of vaccination was verified to be caused by wild measles virus but non-China vaccine strain.
