摘要
目的对接种麻疹减毒活疫苗(Measles Attenuated Live Vaccine,MV)后第9d出现的一例麻疹样病例标本,进行疫苗株和野毒株麻疹病毒鉴定。方法采用两种逆转录-聚合酶链反应-限制性片段长度多态性分析(Reverse Transcription-Polymerase Chain Reaction-Restriction Fragment Length Polymorphism,RT-PCR-RFLP)方法,检测麻疹样病例咽拭子标本。结果应用H1基因型麻疹野病毒RT-PCR-RFLP方法,对该病例咽拭子标本进行鉴定,其RT-PCR阳性产物经SalI酶切后,被切成两个片段,分别为408个和159个核苷酸,显示该病例是由H1基因型麻疹野病毒引起的。应用中国MV株RT-PCR-RFLP方法对该病例咽拭子标本进行鉴定,其RT-PCR产物不能被AflⅡ酶切,仍呈单一条带,显示该病例是由非MV株病毒引起的。结论接种MV第9d后出现的麻疹样病例,被证实为H1基因型麻疹野病毒引起,而非MV株病毒引起。
Objective To distinguish measles vaccine strain and wild strain from a measles case after measles vaccination at the 9th day.Method To apply 2 kinds of methods of Reverse TranscriptionPolymerase Chain Reaction-Restriction Fragment Length Polymorphism(RT-PCR-RFLP) to detect NPS specimen of measles.Results The RT-PCR-RFLP method for H1 genotyping was used for identifying this case,and its RT-PCR products were cut into 2 fragments of 408 bp and 159 bp,the results showed that this case was caused by H1 genotype.The RT-PCR-RFLP method for vaccine strain identification was used for identifying this case,and its RT-PCR products couldn't be cut by AflⅡ enzyme and showed a single band.The results showed that this case was caused by non-China vaccine strain.Conclusions The case who got measles after 9 days of vaccination was verified to be caused by wild measles virus but non-China vaccine strain.
