复方血竭对烫伤大鼠肠黏膜屏障的实验研究 被引量:2

Experimental study of resina draconis in protecting intestinal mucous membrane barrier in burned rats

摘要 [目的]探讨复方血竭对烫伤大鼠肠黏膜屏障的影响。[方法]雄性Wistar大鼠70只,随机分为无烫伤组10只,烫伤组及烫伤后使用复方血竭组(血竭组)各30只。用沸水制成大鼠背部35%Ⅱ度烫伤,血竭组于烫伤后立即、4 h、8 h用复方血竭20 ml/kg灌胃,烫伤组和无烫伤组口服等量0.85%氯化钠。烫伤组、血竭组分别于烫伤后8、12、24 h处死动物,无烫伤组8h全部处死。大鼠FITC-D灌胃测定肠黏膜通透性,穿刺腹主动脉测血浆白细胞介素(IL)-1β、IL-4、IL-6、IL-10的水平。[结果]烫伤后大鼠门静脉FITC-D标记的葡聚糖及血浆IL-1β、IL-6与无烫伤组相比均显著升高(P<0.01)。使用复方血竭后,门静脉FITC-D标记的葡聚糖及血浆IL-1β、IL-6均明显下降,血竭组IL-1β、IL-6水平较烫伤组明显降低,IL-4、IL-10水平明显升高(P<0.05或P<0.01)。[结论]烫伤可导致大鼠肠黏膜应激损伤,肠黏膜通透性增加,血浆IL-1β、IL-6升高;复方血竭可以保护应激状态下的肠黏膜屏障,抑制炎症因子IL-1β、IL-6的产生,上调IL-4和IL-10等抑炎性因子,纠正异常的免疫功能,降低免疫细胞对炎症的反应性,从而有利于炎症的消除及组织的修复,减轻全身炎症反应,保护脏器功能。 [Objective]To investigate the effect of resina draconis on intestinal mucous membrane barrier of burned rats.[Methods]Seventy male Wistar rats(weight of 200-250g)were randomly divided into three groups:normal group(n=10),burned group(n= 30)and treatment group treated with resina draconis(n= 30).The burned model was subjected to a 35% TBSA second degree scald.The treatment group was administered with resina draconis immediately,4h and 8h post burn,and the other two groups with the same volume 0.9% sodium chloride.Rats of burned group and treatment group were killed at 8h,12h and 24h post burn respectively.The rats of the normal group were killed at 8h.FITC-D in portal and IL-1β,IL-4,IL-6 and IL-10 in plasma were measured.[Results]In comparison with the burned group,the treatment group had lower portal FITC-D levels,plasma IL-1β and IL-6 levels,increased IL-4 and IL-10(P〈0.01 or P〈0.05).[Conclusion]After thermal injury IL-1β,IL-6 levels in plasma increased as a result of intestinal mucosal barrier injury.Resina draconis was effective in protecting rats against burn stress,which may result from inducing IL-1β and IL-4 in intestinal mucosa.
