实验性脑出血灶周Caspase-3和Bcl-2的表达及意义

摘要 目的 观察脑出血后不同时间出血灶周凋亡细胞、Caspase-3、Bcl-2的表达.方法 健康雄性Wistar大鼠48只随机分成假手术组和脑出血组,大鼠尾壳核区注入自体非抗凝动脉血建立脑出血模型,采用TUNEL法检测凋亡细胞,免疫组织化学方法观察术后不同时间点Caspase-3和Bcl-2的动态变化.结果 脑出血组大鼠出血灶周凋亡细胞及Caspase-3、Bcl-2的表达较假手术组显著增强(p〈0.01).结论 脑出血后血肿周围神经细胞损伤有凋亡机制参与,且与Caspase-3和Bcl-2蛋白表达有关. Objective To observe neuron apoptosis, Caspase-3 and Bcl-2 in rat models of experimental ICH at different times. Methods Forty-eight rats were randomly divided into control and ICH groups.ICH rat model was made by putting fresh quantitative autoiogous blood into the left caudate nucleus of rat.Terminal deoxynucleotidyl transferase mediated deoxyuridine triphosphate-bioti in situ nick end-labeling(TUNEL) was used to detect apotosis cells and the activation of Caspase-3 and Bcl-2 was measured with immunohistochemistery. Results The expression of apotosis cells,Caspase-3 and Bcl-2 in ICH group was higher than the control group(P〈0.01). Conclusion Apotosis is a pattern of death after intracerebral hemorrhage.Apoptosis has relationship with Caspase-3 and Bcl-2 expression.
