摘要
目的 ATM(ataxia-telangiectasia mutant)蛋白与细胞放射敏感性密切相关。该研究探讨ATM蛋白在不同放射敏感性的大肠癌细胞系HM7和HT29中的表达。方法应用免疫荧光标记技术,对HM7和HT29中的ATM蛋白进行激光扫描共聚焦显微镜(LSCM)分析。结果 ATM蛋白定位于细胞核及胞浆中,并以细胞核为主,且HM7细胞核中ATM蛋白阳性细胞的荧光强度较HT29强;半定量分析,HM7中ATM蛋白表达的积分吸光度值为8×106,HT29中为3×106。结论 ATM蛋白的表达在HM7和HT29中有差别,这种差别可能与它们所具有的不同放射敏感性有关。
【Objective】 To determine the expression of ATM protein in two colon cancer cell lines with different radiosensitivity.【Methods】 Two colon cancer cell lines with different radiosensitivity,HM7 and HT29,were established.The localization and quantity of ATM protein were analyzed by fluorescence immunohistochemical method and laser scanning confocal microscope(LSCM).【Results】 ATM protein was located in both nucleus and cytoplasm,it was more strongly expressed in nuclei of HM7 than that in nuclei of HT29.【Conclusion】 The expressions levels of ATM protein differ in HM7 and HT29.The variance may be related to their different radiosensitivity.
