β_1整合素与卵巢癌细胞凋亡关系的实验研究

摘要 目的研究β1整合素及纤维黏连蛋白在人卵巢癌HO-8910细胞增殖、凋亡中的作用及其意义。方法以培养在包被纤维黏连蛋白底物上的人卵巢癌HO-8910细胞为靶细胞,用不同浓度抗β1整合素单克隆抗体阻断β1整合素与纤维黏连蛋白的相互作用。用MTT法测定细胞增殖抑制率;吖啶橙(acridine orange,AO)染色观察细胞凋亡的形态学变化;流式细胞仪检测细胞周期分布及细胞凋亡率。结果抗β1整合素单克隆抗体能抑制人卵巢癌HO-8910细胞的增殖,并有明显的时间和剂量依赖性(P<0.05)。荧光显微镜观察到细胞凋亡的形态学改变;流式细胞术检测结果显示实验组S期上升,细胞凋亡率上升。结论抗β1整合素单克隆抗体可抑制卵巢癌细胞的增殖,β1整合素抗体与卵巢癌细胞凋亡密切相关,阻断β1整合素与其特意配体纤维黏连蛋白的相互作用可诱导卵巢癌细胞凋亡。 【Objective】To investigate the effects of β1 integrin and fibronectin on multiplication and apoptosis of ovarian carcinoma cell line Ho-8910.【Methods】Human ovarian carcinoma cell line Ho-8910 cultured on the peridishes coated by fibronectin in vitro was treated with monoclonal antibody against β1 subunit at different concentrations to block the interaction between β1 integrin and fibronectin.The multiplication of ovarian carcinoma cells was determined by methyl thiazolyl tetrazoliurm(MTT) method.Acridine orange(AO) were used to assay cell apoptosis and ultrastruccture changes.Cell cycle,apoptotic percentage were detected by flow cytometry.【Results】Ovarian carcinoma cell line Ho-8910 was induced significantly by monoclonal antibody againstβ1 subunit.Within some limits,the effect was time and dose dependent.MTT showed that with the concentration of monoclonal antibody against β1 subunit increasing,it inhibited the growth of ovarian carcinoma cells significantly.Apoptosis morphology was observed in β1 integrin groups.FCM showed that the proportion of S phase and the apoptotic rate were increased significantly in experiment array.【Conclusion】Ovarian carcinoma cell apoptosis may be related to β1 integrin.Cell survival was increased by the interaction between β1 integrin and fibronectin.Cell apoptosis was induced by blocking the interaction.
