人类巨细胞病毒感染与血脂代谢关键酶的关系 被引量:5

摘要 目的研究人类巨细胞病毒(HCMV)感染与肝脂酶(HL)、脂蛋白脂肪酶(LPL)及卵磷脂胆固醇酰基转移酶(LCAT)3个血脂代谢关键酶的含量或相对活性的关系,初步探讨HCMV致动脉粥样硬化的机制。方法利用ELISA技术检测研究对象HCMV-IgG、HL及LPL,用自身底物法检测LCAT的相对活性并对数据进行分析。结果 HCMV-IgG阳性组HL含量为(170.65±89.14)μg/mL,LCAT相对活性为(30.85±19.72)%,均明显低于HCMV-IgG阴性组(218.74±104.58)μg/mL及(39.16±22.44)%,t值分别为2.2316和2.2633,均P<0.05。HCMV-IgG阳性组与HCMV-IgG阴性组LPL差异无显著性(t=1.4834,P>0.05)。HCMV-IgG抗体指数在20.1以上或10.1以上者,血浆HL含量或LCAT相对活性均分别明显低于HCMV-IgG抗体指数20.0或10.0以下各组;相关性分析表明,HCMV-IgG抗体指数值与HL含量及LCAT相对活性存在明显的负相关关系,相关系数分别为-0.4612和-0.5541(均P<0.05)。结论 HCMV感染可通过改变宿主基因表达致HL及LCAT含量或相对活性下降,从而导致宿主脂代谢异常,最终导致动脉粥样硬化的形成。 【Objective】To investigate the relationship between HCMV infection and content of hepatic lipase(HL),lipoprotein lipase(LPL) and the relative activity of lecithin-cholesterol acyltransferase(LCAT),the key enzymes of blood fat metabolism,and to clarify the biochemical mechanisms of atherosclerosis induced by human cytomegalovirus preliminary.【Methods】The detection of HCMV-IgG,HL and LPL by ELISA and the relative activity of lecithin-cholesterol acyltransferase by self-substrate method were carried out,and the data were analyzed.【Results】In HCMV-IgG positive group,the content of HL [(170.65±89.14) μg/mL] and the relative activity of LCAT [(30.85±19.72)%] was much lower than that in HCMV-IgG negative group [(218.74±104.58) μg/mL] and(39.16±22.44)%,respectively.T value was 2.2316,2.2633,and P value was all less than 0.05,while no obvious difference of the contents of LPL between HCMV-IgG positive group and the negative one was observed(t =1.4834,P 0.05).HL contents or LCAT relative activity in groups with HCMV-IgG antibody index more than 20.1 or 10.1 were much lower than that in groups with HCMV-IgG antibody index less than 20.0 or 10.0,respectively.An obvious negative correlation between HCMV-IgG antibody index and the content of HL or the relative activity of LCAT was observed(the correlation coefficient was-0.4612 and-0.5541 respectively,and the P value was all less than 0.05).【Conclusion】HCMV infection may down-regulate the expression of HL and LCAT,resulting in abnormal metabolism of blood fat and formation of atherosclerosis eventually.
