摘要
目的:建立消风止痒颗粒中防风的含量测定方法。方法:采用Agilent zorbax SB-C18(4.6mm×250mm,5μm)色谱柱,以甲醇-水进行梯度洗脱(0~25min时30%甲醇,26~50min时40%甲醇),流速1.0ml/min,检测波长为254nm,柱温30℃。结果:防风中两种主要成分升麻素苷和5-O-甲基维斯阿米醇苷的线性范围分别为0.02007~0.4014μg(r=0.9999)以及0.02113~0.4226μg(r=0.9999),平均回收率(n=6)分别为97.6%(RSD=1.2%)以及97.8%(RSD=1.2%)。结论:所建立的HPLC法可用于测定消风止痒颗粒中防风的含量。
Objective: To develop a RP-HPLC method for the determination of main active components of Saposh- nikovia divaricata (Turcz.) Schischk. in Xiaofengzhiyang granules. Methods: An Agilent zorbax SB-C18 column (4.6mm×250mm, 5μm)was used. The mobile phase consisted of methanol and water(0-25min,30%methanol; 26- 50min,40%methanol) at the flow rate of 1.0ml/min, the detecting wavelength was set at 254nm and the temperature of column was 30℃. Results: Prim-O-glucosylcimifugin and 4'-O-glucopyranosyl-5-O-methylvisammino showed good linearity (r=0.9999) in the range of 0.02007-0.4014μg and 0.02113 -0.4226μg, respectively. The average recovery was 97.6% and 97.8%(n=6), respectively. Conclusion: The developed method can be applied for quality control of Saposhnikovia divaricata(Turcz.) Schischk. in Xiaofengzhiyang granules.
