鸦油乳对前列腺增生动物模型的影响及机理研究 被引量:14

An experimental study of the therapeutic effect of brucea javanica oil emulsion on benign prostatic hyperplasia

摘要 目的 探讨鸦胆子油静脉乳剂( 简称鸦油乳) 对前列腺增生的影响及作用机理。 方法 将SD大鼠去势7 天后皮下注射睾酮4mg/kg ,同时腹腔注射10 % 鸦胆子油乳5mg/kg 。于给药10 、20 、30 天时处死大鼠,观察前列腺重量,通过光镜及电镜观察前列腺组织细胞结构变化。通过免疫组化法检测PCNA 和bcl2 蛋白表达的改变。 结果 给药20 天后鸦油乳组大鼠前列腺重量明显低于睾酮组,电镜证实鸦油乳可破坏前列腺上皮细胞的细胞膜、粗面内质网和基膜等,使细胞变性坏死。免疫组化法证实在给药10 天时,鸦油乳组PCNA 指数明显低于睾酮组,而bcl2 蛋白表达各组间差异无显著性。 结论 鸦油乳可通过破坏细胞膜性结构抑制细胞增生,从而发挥治疗前列腺增生的作用。 Objective To study of the effect of brucea javanica oil emulsion (BJOE) on benign prostatic hyperplasia in animal models. Methods Castrated SD rats were administered exogenous testosterone and at the same time 10% BJOE intraperitoneally.The rats were decapitated at 10,20 and 30 days respectively and the weight of prostate,light microscopy and electron microscopy were undertaken.PCNA index and bcl 2 protein were analyzed by means of immunohistochemical technique. Results At the 20th day after BJOE administration,the weight of the prostate was significantly less than the controls.Significant suppression of growth was noted in the BJOE groups with the rough endoplasmic reticulum and cell membrane ruined.Remarkable decrease of PCNA index was also observed but bcl 2 protein showed no significant change at the 10th day. Conclusions BJOE would inhibit cell hyperplasia of prostate by destroying the cell membranous system.The results of the experimental study provided a strong theoretical basis for the clinical use of BJOE in the treatment of human BPH.
